2 CHƢƠNG 1: TỔNG QUAN
1.1. HISTON DEACETYLASE (HDAC)
Tất cả bộ gen của ngƣời đƣợc gói vào trong nhiễm sắc thể (NST), một phức
hợp đại phân tử (protein-AND) có cấu trúc động. NST bao gồm các proteinhiston
và protein không phải histon. Có năm loại protein histon chính: H2A, H2B, H3, H4
và H1[21]. Đơn vị cấu trúc cơ bản của NST là các nucleosom có đƣờng kính 100Å.
Một nucleosom điển hình bao gồm một octamer hình đĩa của 4 cặp histon (2H2A,
2H2B, 2H3, 2H4) đƣợc quấn quanh bởi 146 cặp nucleotid[1,4,57] (hình 1.1)

1.1.1. Khái niệm về histon deacetylase
HDAC là một nhóm các enzym đƣợc phát hiện ở nhiều loại sinh vật nhƣ vi
khuẩn, nấm, thực vật và động vật;nó có tác dụng xúc tác quá trình loại bỏ nhóm
acetyl từ ε-N-acetyl lysin amino acid của histon.
Histon acetyltransferase (HAT): là enzym acetyl hoá nhóm ε-NH
2
trong gốc
lysin (đầu N tận) của histon,làm trung hoà điện tích dƣơng trên lysin, vì vậy giảm
khả năng tƣơng tác của histon (tích điện dƣơng) với ADN (tích điện âm), tạo ra cấu
trúc mở của chromatin. Vì vậy, sự acetyl hoá histon tạo điều kiện cho quá trình
phiên mã, dịch mã xảy ra.
HDAC có tác dụng đối lập với histon acetyltransferase (HAT) - HDAC xúc
tác loại bỏ nhóm acetyl từ lysin ở đầu Ntận của histon, làm đóng xoắn chromatin, do

Các nghiên cứu cho thấy mức độ acetyl hóa ảnh hƣởng trực tiếp tới chức năng
cơ bản của tế bào nhƣ quá trình phiên mã, tính di động, biệt hóa, chết theo chu kỳ
của tế bào. Sự sai khác trong acetyl hóa do sự gián đoạn hoạt động của các HAT
hoặc hoạt động mạnh bất thƣờng của các HDAC đã đƣợc chứng minh là liên quan
tới sự hình thành và sự tiến triển của ung thƣ [25,60].
HDAC xúc tác loại bỏ nhóm acetyl từ histon và protein không phải histon
(p53, hsp90 hay tubulin), cụ thể HDAC xúc tác cho phản ứng deacetyl hóa lysin tạo
ra nhóm NH
3
+
ở phần đuôi của histon (hình 1.2) [69], làm tăng sự tích điện dƣơng
trên đầu N của histon và đóng xoắn NST, kết quả là ức chế quá trình phiên mã do
ngăn cản các nhân tố sao mã tiến đến đích của chúng trên AND. Các sai lệch của
quá trình phiên mã là một trong những nguyên nhân dẫn tới sự hình thành khối u
[4].
HAT acetyl hóa histon làm trung hòa cực dƣơng trên lysin, cụ thể HAT xúc
tác cho phản ứng acetyl hóa nhóm ε-NH
2
trong phân tử lysin ở phần đuôi của histon
5 làm hoạt hóa quá trình phiên mã nhờ vào việc tháo xoắn cấu trúc của nucleosom, do
đó giúp cho các yếu tố sao mã dễ dàng tiếp cận AND.
Vậy sự acetyl hóa và deacetyl hóa histon trong NST đóng vai trò quan trọng
trong quá trình biểu hiện gen. Việc mất cân bằng hoạt động giữa HAT và HDAC có
thể dẫn đến những bất thƣờng biểu hiện gen và do đó dẫn đến ung thƣ
[24,34,39,45,48].
Mối liên quan giữa hoạt động của HDAC và sự tạo thành các khối u đƣợc thể

là coenzym của HDAC nằm ở gần cuối túi thân dầu. Đây là phần
tham gia liên kết mạnh nhất với phần đuôi lysin bằng các liên kết phối trí. Thông
thƣờng các chất ức chế liên kết càng mạnh với ion Zn
2+
thì hoạt tính sinh học càng
mạnh.
-Kênh enzym là nơi chứa đựng và tham gia tạo liên kết Van der Waals với cơ
chất. Kênh này có cấu trúc dạng túi, toàn kênh đƣợc cấu tạo bởi các acid amin thân
dầu, đặc biệt là các acid amin có nhân thơm nhƣ: phenylalanin, tyrosin, prolin,
histidin. Cấu trúc kênh khá linh động, có thể thay đổi kích thƣớc để phù hợp với cơ
chất trong phản ứng deacetyl. Ở HDAC nhóm I kênh này có chiều sâu khoảng 11Å,
ở gần cuối kênh đƣờng kính của nó khoảng 7Å và đƣợc hạn chế bởi hai acid amin
phenylalanin nằm song song với nhau (hình 1.4)

7 1.2. CÁC CHẤT ỨC CHẾ HDAC
1.2.1. Phân loại các chất ức chế HDAC
Ngày nay có rất nhiều chất có tác dụng ức chế HDACđã đƣợc nghiên cứu và
công bố cho thấy tiềm năng trong việc điều trị ung thƣ. Chúng có thể có nguồn gốc
tự nhiên hoặc tổng hợp hóa học. Chất có tác dụng mạnh nhất đƣợc biết cho đến nay
là TSA - một sản phẩm lên men của Streptomyces [48]. Hiện tại có 3 chất ức chế
HDAC đƣợc FDA cấp phép lƣu hành trên thị trƣờng có tác dụng điều trị u lympho
tế bào T dƣới da: SAHA (vorinostat, Zolinza
®
